Personally I love the subject of the anointing. However, in reading through the posts in this thread I think that I would change the question to,
"what is the anointing?"
Many have made references to the Holy Spirit which is true, but there are different terminologies throughout the Bible that we can synonymously say is the Spirit of God...Presence, glory, anointing, Holy Ghost and so forth.
The question then becomes, why is different terminologies use? I would submit the terminology is directly related to the aspect of God being referenced.
Anointing...what is it?
Isa 10:27
27 And it shall come to pass
in that day, that his
burden shall be taken away from off thy shoulder, and
his yoke from off thy neck, and the yoke shall be destroyed
because of the anointing. (KJV)
In the above scripture the Prophet Isaiah defines the anointing. Because of the anointing the devils burdens are taken away and the devils yokes are destroyed. But notice Isaiah said, "...in that day..." Let's find the "day" of fulfillment of the anointing.
Luke 4:17-21
17 And there was delivered unto him the book of the prophet Esaias. And when he had opened the book, he found the place where it was written,
18 The Spirit of the Lord is upon me, because he hath
anointed me to preach the gospel to the poor; he hath sent me to heal the brokenhearted, to preach deliverance to the captives, and recovering of sight to the blind, to set at liberty them that are bruised,
19 To preach the acceptable year of the Lord.
20 And he closed the book, and he gave it again to the minister, and sat down. And the eyes of all them that were in the synagogue were fastened on him.
21 And he began to say unto them,
This day is this scripture fulfilled in your ears. (KJV)
While I understand that the passage from Isaiah that Jesus was a different passage, the prophesy of Isaiah in about the anointing was fulfilled in this day also. Notice what Jesus said He was anointed to do...remove burdens and destroy yokes.

While I realize that what Jesus was reading was a different section of Isaiah, the prophesy of 10:27 was also fulfilled here. Notice what Jesus was anointed to do - remove burdens and destroy yokes. Poverty, sickness, brokenheartedness, addictions, blindess, and bondage are all forms of burdens and yokes.
Many talk about having the Holy Ghost, but I have seen many Spirit filled people just as bound as non-believers. There is a difference between being Spirit filled and having the power to remove burdens and destroy yokes.
I have seen worship leaders who while leading worship the Spirit moves in such a way that people are healed, filled with the Spirit, and set free. I would say they are "anointed" in music. I have seen other worship leaders that a good, but they lack the anointing.
I have seen preachers that can preach a good word and others who through their preaching can bring freedom into peoples lives.
The manifestation of the anointing does something...in my opinion.
